Abstract
Malaysia has the highest prevalence of obesity in the South East Asia region (Cheong, 2014). There are only a few anti obesity drugs available to treat obesity, however, most of them have a lot unwanted effects (Kang & Park, 2012). According to Lim et al. (2014), germinated brown rice (GBR) is suggested as an alternative in reducing lipid accumulation in 3T3-Ll adipocytes. Thus, this study might provide further evidence of GBR that might be a good alternative in reducing lipid accumulation in adipocytes. This study aims to determine the optimal concentration of GBR methanolic extraction on reducing 3T3-Ll cells differentiation and to study the effect of methanolic GBR extract in 3T3-Ll adipocytes cells. The germinated brown rice was extracted with methanol using soxhlet apparatus. Both 3T3-Ll adipocytes and WRL68 cells were treated with five different concentration of BR (62.5 µg/ml, 125 µg/ml, 250 µg/rnl, 500 µg/ml, 1000 µg/ml). The cells viability and lipid accumulation were identified by using MTS assay and Oil Red O staining. This study revealed that the IC50 (62.5 µg/ml ) for adipocytes cells is higher IC50 (250 µg/ml) for WRL68. Besides that, the lipid accumulation in adipocytes reduced when treated with the GBR extract.
