Abstract
The incidence of dengue virus infection is dramatically increasing in recent decades. It is endemic in more than 100 countries and about 2.5 billion people are at risk of dengue virus infection. It is important to identify and isolate the specific cell surface receptor for dengue virus to understand its mechanism. There are few new receptors that have been reported have a role in the dengue virus entry mechanism and one of it is DC-SIGN. The aim of this study is to identify and amplify the homology arms of DC-SIGN gene. The homology arms produced can be used by future researchers to produce the gene knockout for DC-SIGN gene. To design primers that are specific to DC-SIGN gene, we used the ENSEMBL software, RepeatMasker software and PrimerBlast software. The amplification of homology arms was done by PCR methods together with 0.25 µL of DNA Taq Polymerase. The size of the PCR product is 1000 bp. All PCR products were analysed with 1.5% agarose gel electrophoresis method by running it together with 1000 bp DNA Ladder marker. The result shows a successful amplification of homology arms for DC-SIGN gene. The amplification of homology arms of DC-SIGN gene can lead to new discoveries in dengue research. The production of gene knockout of DC-SIGN gene can give a better understanding of the mechanism of entry of dengue virus.
